![]() In The Films of Tom Hanks, authors Lee Pfeiffer and Michael Lewis describe one of the ways Ephron helped bridge the gap between Sam and Annie, "Ryan was going in one door in Baltimore while Hanks was going through another in Seattle. It may be easy to understand Sam Baldwin, but it was more difficult for Ephron to build a love story between two characters before they ever even meet. In describing his character, Hanks stated, "The guy is enmeshed in grieving, and no one has to work hard in buying that attractive premise, as opposed to a guy who gets off the airline and picks up the wrong suitcase and it's full of uranium." Hanks liked that the role was more serious than the parts he'd played in his earlier movies. Tom Hanks initially turned down the role of Sam Baldwin thinking he wasn't right for the part, but later changed his mind after Nora Ephron rewrote the script. She received her third nomination for co-writing the script for Sleepless in Seattle. Ephron received Academy Award nominations for her screenplays for Silkwood (1983) and When Harry Met Sally (1989). She then went on to write the semi-autobiographical novel Heartburn, which was made into a film in 1986. Nora Ephron began her career as a journalist for The New York Post. The play was later made into the film Take Her, She's Mine (1963) with Sandra Dee and Jimmy Stewart. The Ephrons also wrote a play loosely based on letters Nora sent them from Wellesley College. Their work includes the films, Carousel (1956), Daddy Long Legs (1955), and Desk Set (1957). You want to be in love in a movie." Nora Ephron thought this was one of the intriguing aspects of Sleepless in Seattle and noted that "It had all these weird, wonderful ideas to play with, including all this stuff about what the movies do to your brain and how so many of our notions about romance are based on the movies we've seen."Įphron learned about movies at an early age thanks to her parents, screenwriters Henry and Phoebe Ephron. Annie comments, "Now that was when people KNEW how to be in love." Becky replies, "That's your problem! You don't want to be in love. ![]() The mice could father pups again 4-6 weeks after they stopped receiving the compound.Īccording to Georg, YCT529 will begin testing in human clinical trials in the third or fourth quarter of 2022. When given orally to male mice for 4 weeks, YCT529 dramatically reduced sperm counts and was 99% effective in preventing pregnancy, without any observable side effects. They identified a compound, which was named YCT529, that inhibited RAR-α almost 500 times more potently than it did RAR-β and -γ. With this information, they designed and synthesized approximately 100 compounds and evaluated their ability to selectively inhibit RAR-α in cells. So the researchers closely examined crystal structures of RAR-α, -β and -γ bound to retinoic acid, identifying structural differences in the ways the three receptors bind to their common ligand. ![]() Other scientists have developed an oral compound that inhibits all three members of the RAR family (RAR-α, -β and -γ) and causes reversible sterility in male mice, but Georg’s team and their reproductive biology collaborators wanted to find a drug that was specific for RAR-α and therefore less likely to cause side effects. Knocking out the RAR-α gene in male mice makes them sterile, without any obvious side effects. This protein is one of a family of three nuclear receptors that bind retinoic acid, a form of vitamin A that plays important roles in cell growth, differentiation (including sperm formation) and embryonic development. To develop their non-hormonal male contraceptive, the researchers targeted a protein called the retinoic acid receptor alpha (RAR-α). ![]() “We wanted to develop a non-hormonal male contraceptive to avoid these side effects,” says Noman, a graduate student in the lab of Gunda Georg, Ph.D., at the University of Minnesota. Most compounds currently undergoing clinical trials target the male sex hormone testosterone, which could lead to side effects such as weight gain, depression and increased low-density lipoprotein (known as LDL) cholesterol levels. “Scientists have been trying for decades to develop an effective male oral contraceptive, but there are still no approved pills on the market,” says Md Abdullah Al Noman, who is presenting the work at the meeting. Therefore, men need an effective, long-lasting but reversible contraceptive, similar to the birth control pill for women. Although vasectomies can sometimes be reversed, the reversal surgery is expensive and not always successful. In contrast, vasectomy - a surgical procedure - is considered a permanent form of male sterilization. However, condoms are single-use only and prone to failure. The meeting features more than 12,000 presentations on a wide range of science topics.Ĭurrently, men have only two effective options for birth control: male condoms and vasectomy. ACS Spring 2022 is a hybrid meeting being held virtually and in-person March 20-24, with on-demand access available March 21-April 8. The researchers will present their results today at the spring meeting of the American Chemical Society (ACS). Today, scientists report a non-hormonal male contraceptive that effectively prevents pregnancy in mice, without obvious side effects. But men’s birth control options - and, therefore, responsibilities - could soon be expanding. To avoid getting pregnant, you do not have sex on or around the days you are fertile, or you use a barrier method of birth control.SAN DIEGO, MaWomen have many choices for birth control, ranging from pills to patches to intrauterine devices, and partly as a result, they bear most of the burden of preventing pregnancy. These methods focus on knowing which days of the month you are able to get pregnant (fertile), often based on basal body temperature and cervical mucus. You put these products in the vagina right before sex. Vaginal pH regulator gel (Phexxi) stops sperm from moving, so they can't get to an egg to fertilize it. Spermicide is a type of contraceptive that kills sperm or stops it from moving. These are nonhormonal options for birth control. Examples include tubal ligation for women and vasectomy for men. ![]() This is a permanent method of birth control. These are considered long-acting methods because they last for three to 10 years after insertion - depending on the device - or until you decide to have the device removed. Examples include the copper IUD (ParaGard), the hormonal IUD (Mirena, Skyla, Kyleena, others) and the contraceptive implant (Nexplanon). These are considered short-acting methods because you have to remember to use them on a daily, weekly or monthly basis. Deciding whether pitting for fresh rubber during an enduro to salvage a race objective would’ve added an interesting layer of strategy not being able to do so is nonsense, really. It’s a bizarre omission, and it’s one that quietly undermines a lot of what Autosport does right. You can bring a little logic to the tyre wear issue by manually increasing the length of your endurance races but that only serves to highlight Autosport’s most egregious sin: no pit stops. ![]() It’s strange because tyre wear, which is only a factor in the Endurance category, is scaled to the length of the race. They look the part, but the default length of these races is just eight minutes. The AI makes mistakes, although sometimes they make the sames ones even after multiple race restarts.Įndurance is Autosport’s most confusing category. Drifting feels a bit more forgiving (and more fun) than it did in Grid 2 but the Time Trials here aren’t really that pulse-raising they honestly just feel like a qualifying session for a race that never comes. The Tuner category is probably the weakest of the five. Autosport may be Grid going back to its roots, but it still favours fun over unflinching accuracy. Open Wheel racing is not unlike Codemasters’ own F1 series, although it’s nowhere near as brutally unforgiving as the latter can be. The default length for the races errs on the short side, but there’s an option to multiply race distances by up to five should you find yourself yearning for longer contests. Real-life circuits are the focus here (there are 13, compared to Grid 2’s five, and most feature several routes), and Codemasters has reinserted practice periods and qualifying rounds. Autosport lacks the real-world championships of its Race Driver ancestors, but the aggressive doorhandle-to-doorhandle nature of touring car racing is extremely well-emulated. The Touring category is where Autosport most resembles the typical Codemasters circuit racers of old. Autosport’s prestige team, Ravenwest, demands top results but shells out plenty of XP and allows you to adjust a variety of tuning options at your own discretion. Less bold teams have easily achievable success targets but modest XP rewards, and you’ll usually find you’re not able to tune your car a great deal, if at all. The better you perform in each category the more offers you’ll receive from rival teams. ![]() Autosport doesn’t care that I’d unlocked all of its touring car events before I’d even started scratching the surface of its Street events, nor will it care if you do the exact opposite. It was at least two or three sessions before I even competed in my first Street race. As a touring car fan, I found myself exclusively racing tin-tops for many hours upon first booting up. You need to play them all eventually if you want to meet the minimum requirements to unlock the special Grid Grand Slam events (which string together a series of races from across all five types of racing), but outside of that how you progress through the races on offer is determined entirely by you. ![]() In Autosport, progression is divided across five categories you can play through as you desire: Touring, Endurance, Open Wheel, Tuner, and Street.
